A study to compare weekly or event-based use of doxycycline to prevent sexually transmitted infections in men who have sex with men and transgender women using HIV prevention medication.

What is this study about?

This study focuses on the prevention of Sexually Transmitted Infections, which are infections passed from one person to another through sexual contact. Specifically, the research looks at Neisseria gonorrhoeae, which causes gonorrhea, Chlamydia trachomatis, which causes chlamydia, and Treponema pallidum, the bacteria that causes syphilis. The study involves individuals who are already using PrEP, a medication taken to prevent HIV infection.

The purpose of the study is to compare two different ways of using doxycycline to prevent these infections after potential exposure. One method involves taking the medication every week, while the other method involves taking the medication only after specific events occur. Participants will be assigned to one of these two groups to see which schedule is more effective at reducing the number of new infections.

Who Can Join the Study?

  • You must be able and willing to agree to the study by signing a consent form (a document explaining the study details) in either Dutch or English and follow all study rules.
  • You must be 18 years of age or older.
  • You must have been assigned male sex at birth.
  • You must identify as a gay man, a bisexual man, another man who has sex with men, or a transgender woman (a person whose gender identity is different from the sex they were assigned at birth).
  • You must be currently receiving PrEP care (a daily medication taken to prevent HIV, a virus that can weaken the immune system).
  • You must test negative for HIV (meaning you do not have the virus).
  • You must report having had anal sex without a condom with at least one partner who is not your regular partner in the last year.

Who Cannot Join the Study?

  • You cannot join the study if you have an allergy (an extreme reaction to a substance) to doxycycline or any other ingredients used in the study medicine.
  • You cannot participate if you are currently taking other medicines that might react poorly with doxycycline.
  • You cannot join if there is any medical reason listed in the official product information that says you should not take doxycycline.

Investigated drugs:

Doxycycline is an antibiotic used in this study to prevent certain sexually transmitted infections after a potential exposure.

Gonorrhoea – This is a bacterial infection that primarily affects the mucous membranes of the reproductive tract. It is commonly spread through sexual contact. The infection can cause inflammation and discharge in the genital area. In some cases, it may also affect the throat or rectum. If left unchecked, the infection can spread further into the reproductive organs.

Chlamydia – This infection is caused by a specific type of bacteria that targets the reproductive system. It is typically transmitted through sexual activity. Many individuals do not show any obvious signs of the infection initially. As it progresses, it can cause irritation and discharge in the genital tract. The bacteria can also infect other parts of the body like the rectum or throat.

Syphilis – This is a bacterial infection that progresses through several distinct stages. It often begins with a small, painless sore at the site where the bacteria entered the body. Following this initial stage, the infection can move into a period where rashes appear on different parts of the body. If it continues to advance, the bacteria can spread to internal organs and tissues. The progression is typically slow and can affect various systems in the body over time.
