What is timolol maleate?

Timolol maleate PH. EUR. is a chemically synthesised substance, classified as a beta-blocker under ATC code S01ED01.[1] In the trial registry it appears under the product names Timolol 0,5 % AT - 1 A Pharma, 5 mg/ml Augentropfen, Lösung and Timolol Micro Labs 5 mg/ml Augentropfen, Losung, authorised as a medicine in Germany.[1] The NHS dm+d browser lists a 0.25% eye drops formulation containing timolol maleate as the active ingredient.[2]

How does timolol maleate work?

Timolol is a beta-blocker. Beta-blockers act on beta-adrenergic receptors in the body, and blocking these receptors can affect processes such as heart rate and blood pressure. In the context of this trial, timolol is used topically as part of dermatologic management, but the registry record does not describe its mechanism in skin side effects.[1]

Which conditions is it used or studied for?

  • EGFR-mutated advanced or metastatic non-small cell lung cancer (NSCLC) — timolol is studied here not to treat the cancer itself, but as part of managing selected dermatologic adverse events caused by the cancer medicines amivantamab and lazertinib.[1]

How is timolol maleate given?

In the trial, timolol is given as eye drops, solution for topical application or topical administration.[1] The NHS dm+d browser also records timolol 0.25% eye drops as a prescribable ocular product.[2]

What do the clinical trials show?

One Phase 2 trial, 2023-505863-35-00, is authorised and plans to enrol 80 participants with locally advanced or metastatic Stage IIIB/C-IV EGFR-mutated NSCLC.[1] The study compares enhanced dermatologic management against standard-of-care skin management in people receiving first-line amivantamab plus lazertinib. Timolol is one of several topical and oral agents in the enhanced management arm. The primary outcome is the incidence of Grade ≥2 dermatologic adverse events of interest in the first 12 weeks after starting amivantamab and lazertinib.[1]

What side effects are known?

The supplementary source does not list side effects for timolol maleate, so no side-effect information is given here.[2]
