Study of the efficacy and safety of Eloralintide in adults with knee osteoarthritis and overweight or obesity

What is this study about?

This study focuses on individuals living with Osteoarthritis, a condition where the protective tissue at the ends of bones wears down over time, causing pain and stiffness, particularly in the knee. The research also involves people who are classified as Overweight or have Obesity, which refers to having an excessive amount of body fat. The purpose of the study is to investigate the efficacy and safety of a medication called Eloralintide, also known by its code LY3841136.

Participants in the study will receive either Eloralintide or a placebo through a subcutaneous injection, which is a method of delivering medication into the fatty tissue just under the skin. The medication is administered once every week using a pre-filled syringe containing a solution for injection. During the course of the study, the changes in body weight and knee pain levels will be monitored over a period of time.

Who Can Join the Study?

  • You must have a body mass index (BMI), which is a way to measure if your weight is healthy for your height, of 27 or higher.
  • Your body weight must have been stable, meaning it has not changed by more than 5% in the 90 days before starting the study.
  • You must have previously tried to lose weight through diet, which means changing the types and amounts of food you eat, but were unable to do so.
  • You must have osteoarthritis of the knee, which is a condition where the protective tissue at the ends of your bones wears down over time.
  • In addition to knee osteoarthritis, you must meet at least one of the following: be older than 50 years old, experience morning knee stiffness, which is difficulty moving your knee joints when you first wake up, that lasts about 30 minutes, or feel a crackling or grinding sound in the knee when it moves.

Who Cannot Join the Study?

  • You have an active infection (a disease caused by germs like bacteria) in your knee.
  • You have already had, or are planning to have, surgery (a medical procedure involving cutting the body), an endoscopic procedure (a test where a doctor uses a small camera on a tube to look inside the body), or any medical device used for weight loss.
  • You have diabetes, which is a condition where your blood sugar levels are too high. This includes type 1 diabetes, type 2 diabetes, or any other form of the disease.
  • You have used specific medicines to control your blood sugar (the amount of sugar in your blood) within the last 90 days.
  • You have experienced a serious heart condition (a major problem with how your heart works) within the last 90 days.
  • You have used medicines or alternative remedies (natural or non-traditional treatments) meant to help with weight loss within the last 90 days.

Eloralintide is a medication given as an injection under the skin once a week. It is being studied to see how well it helps reduce knee pain in adults with osteoarthritis and to help with weight management in people who are overweight or living with obesity.

Osteoarthritis – This condition occurs when the protective tissue at the ends of the bones wears down over time. It most commonly affects the joints, such as the knees, hips, and hands. As the tissue thins, the bones may begin to rub against each other directly. This process can lead to joint stiffness and discomfort during movement. The condition typically develops gradually as a person ages.

Overweight or Obesity – This condition involves having an excessive amount of body fat relative to body weight. It occurs when the body takes in more energy from food than it uses for movement and daily functions. Over time, this imbalance leads to the accumulation of fat tissue throughout the body. The condition can affect various parts of the body and may progress if energy intake remains high.
